Abstract

A study to determine the effect of maltose supplement in andromed based semen extender on theviability of buffalo’s epididymal spermatozoa following 12 and 24 hour storage as liquid semen. Epididymalspermatozoa were collected by combined methods of slicing, flushing and tissue pressure. The spermatozoawas then diluted with three different extenders, i.e Andromed only (A) as control, Andromed + 0.2% w/vMaltose (M1) and Andromed + 0.4% w/v Maltose (M2) as treatments. The percentages of progressivelymotil spermatozoa in the liquid semen in the following treatment of A, M1 and M2 were 48.33%; 53.33%and 55% respectively (after 12h of storage), and 45%; 46.67% dan 45% respectively (after 24h of storage).Besides, the percentage of live spermatozoa in A, M1 and M2 after 12h of storage were 70.33%; 72 % and72.33 % and after 24h of storage were 66.33%; 70% and 70.67 %. In conclusion, the addition of 0.2% and0.4% w/v maltose into Andromed extender could maintain the life of the spotted buffalo epididymalspermatozoa after storage for 4°C up to 24 hours.

Abstract

The purpose of this study is to examine the activity of Ambon banana extract in gel formulation on thewound healing process of mice skin (Mus musculus albinus) based on gross and microscopic observations.The wound healing process were observe grossly everyday while the microscopic lesion was observed on the3rd, 5th, 7th, 14th and 21st days after skin incisition. The wounded skin were sampled after the mice wereeuthanized for further microscopic observation. The gross parameters were the existence of blood coagulation,scab formation, wound covering and wound size. The microscopic parameters observed the percentation ofwound reepitelization and the the tickness of wound connective tissues (fibroblast). Gross lesion and thefibroblast thickness were presented descriptively as a qualitative data. Grossly, it was observed that thescab formation was faster in mice treated with the extract of ambon banana than that of negative control.Based on the research the ambon banana extrack in gel can be used in the acceleratiof wound healing.

Abstract

Sixty four samples from female and male local chickens in high and low attitude areas of Bali wereexamined for Leucocytozoon infection. Erytrocyte count, haemoglobin level and packed cell volume (PCV)were examined. Data obtained from this study was analyzed by Wilcoxon Mann Whitney Test. The resultshowed that no significant difference was observed on erytrocyte, haemoglobin and PCV values betweeninfected (2,48 x 106/mm; 9,36g/dl; and 33,62% respectively) and uninfected (2,56x106/mm3; 9,47g/dl and34,28% respectively). A significant difference was however observed on the erythrocyte count between malechickens in higher area (2,60x106/mm3) and female in lower area (2,59x106/mm3). No significant differencewas observed on haemoglobin and PCV between male chickens in higher area (9,35g/dl & 34,62% and9,56g/dl & 34,75% respectively) and female chickens in lower area (9,38g/dl & 32,63% and 9,16g/dl &32,5% rspectively).

Abstract

The aim of this research was to determine the lead concentration in blood and hematological profilesof Sumatran elephants. Eight out of 57 captivated Sumatran elephants in Sebanga Elephant TrainingCenter (ETC) Riau were examined for the lead concentration in the blood and for hematological profiles.The results showed that their lead concentration in blood was 0.3425 – 0.4633 ppm with the average of0.3905 ± 0.373 ppm. Their hemoglobin level was 9.60 – 12.10 g/dl with the average of 10.39 ± 1.20 g/dl.Hematocrit levels was 31 – 47% with the average of 37.50 ± 5.21%. In male, their average levels ofhemoglobin and hematocrit were 10.85 ± 1.28 g/dl and 41.00 ± 4.90%, respectively which were higher thanthose in female (9.93 ± 1.06 g/dl and 34.00 ± 2.53%, respectively). Their lead concentration was higher inthe younger elephant than in the older elephants. Although, the lead concentration in blood of Sumatranelephants was high, it appeared to be not related either with their hemoglobin or hematocrit levels. Therewas, however, relationship between the lead level in blood with the age of elephants.

Abstract

Low productivity of in vitro embryo production at blastosis level appears to be as a result of imperfectoocyte maturation which causes imperfect oocyte growth, and in turn will affect the embryo growth. Inaddition to hormonal factor, growth factor plays significant role in maturation process of oocytes. Asgrowth factor might has a significant role during maturation process, a study was conducted to determinewhether transforming growth factor ? (TGF ?) isolated from oocytes of cumulus complex is required for coculture and in vitro embryo production.. Oocytes were collected from follicles with the diameter of 3-5 mmand > 5 mm. Then the oocytes were cultured for 22 hours at 38.5o with 5% CO2 atmosphere in tissueculture medium (TCM) 199 supplemented with 5 ?g/mg luteonizing hormone (LH), 3% bovine serumalbumin (BSA) 50 ?g/ml gentamycin sulfat and three different levels of TGF ? (12,85 pg/ml, 25,7 pg/mland 38,55 pg/ml). The oocyte maturation and number of apoptosis cells were examined. The result showedthat oocyte maturation in medium supplemented TGF ? at the dose of 38,55 pg/ml was better than in thatat dose of 12,85pg/ml or 25,7 pg/ml. The best maturation was observed at metaphase II stage of oocytedevelopment. No apoptosis was observed during maturation of oocytes. Supplementation of TGF ? at thedose of 38,55 pg/ml in culture medium increased the oocyte maturation without causing a significantapoptosis in vitro.

Abstract

Transportated animals may subject to a variety of physical stimuli including metabolism, crowding,noise, handling, isolation, agitation, and extreme temperature .The aim of this study was to determine thechanges of serum T4 and T3 concentration, during animals transportation. Six adult Bligon buck with bodyweight ranging from 26-30 Kg were used in this study. Two weeks prior to the experiment, the animalswere given anthelmintic Albendazole to eliminate egg worm. All animals were fed standard diet in theirpen at 10% of their body weight per head daily and commercial concentrate also given everyday. Freshwater was provided ad libitum. All animals were transported around village for 16 hours starting from18.00 pm until 10.00 am in open small truck (3 x 2 m); eye contact each others would be possible. Bloodsamples were withdrawn from jugular vein using vacutainer tubes containing heparin into 1.5 mL glasstubes, then centrifuged at 500 g for 15 minutes. Plasma was collected to be stored at –20° C. The bloodwere collected every 4 hours from 8 hours before transportation ( at 10.00 am, 14.00 pm and 18.00 pm)until the time of arriving after transportation at 10.00 am. Plasma was harvested and stored at –20° Cuntil T3 and T4 concentrations were measured using ELISA method (enzyme linked immunosorbentassay) product DRG, Germany. The result showed that transportation of Bligon bucks for 16 hours havean affect on level of T4 only (P<0.05) and not for T3 concentration (P>0.05) due to physical stimuli such ascrowding, heat stress, noise, handling would be discarded so that the metabolic process was stable. Duringtransportation, decreasing of T4 levels indicated conversion of T4 to T3 to form active hormone.

Abstract

A study was conducted to determine the effectiveness of oxytetracycline on the basis of itspharmacokinetic/pharmacodynamic (PK/PD) parameters in the plasma and tissues of broiler following a50 mg/kg single dose of intravenous administration. The male broiler were injected with oxytetracycline attarsal medial venous. The blood, liver, kidney and abdominal muscle were collected at 14 points of timesamplings. Blood was centrifuged to obtain plasma and all tissues were extracted with Mc Ilvine buffer.Drug concentration in samples was determined by High Performance Liquid Chromatography (HPLC)using AOAC standard reference for tetracycline. The pharmacokinetic parameters of oxytetracycline werecalculated using non compartment method. The results were as follows. For plasma: the AUC was 790,615,5 ?g/mL minute , Clearance was 63,242 mL/min/kg bb, Tmax/Cmax was 1 minute/0,7 ?g/mL, T1/2elimination was 6,4, hours, Vd was 37 L/kg. For liver ; AUC was 36418,89, minute , Tmax/Cmax was 16hours/17,15 ?g/g, and T1/2 elimination was 24,5, hours. For kidney; AUC6808,41 ?g/g minute , Tmax/Cmax 1 minute/16,73 ?g/g, , T1/2 elimination was 11,55 hours. For muscle; AUC was 3502 ?g/g minute,Tmax/Cmax was 1 minute/2,58 ?g/g, T1/2 elimination was 167,39 hours. The ratio of AUC/MIC inplasma and tissues appeared to a good pharmacokinetic/pharmacodynamic parameter to determine theeffectiveness of oxytetracycline.

Abstract

The aim of this research was to prepare rabbit Immunoglobulin G as anti-idiotype antibody (Ab2) ofAvian Influenza Virus (AIV) H5N1. A polyclonal antibody was collected from guinea pigs immunized withinactivated AI vaccine H5N1of Legok strain. Antibody of H5N1 AI in serum was detected by Agar gelprecipitation test (AGPT) and an Inhibition Hemmaglutination test (IHT). The highest titre of antibodywas obtained one week after the third immunization. Serum of guinea pigs containing IgG was purifiedusing the Montage Antibody purification kit & spin column with Prosep A media (Millipore). The AI H5N1IgG concentration was 8 mg/ml. AI H5N1 IgG, was then digested with pepsin to obtain F(ab)2 fraction andwas called Ab1. The concentration of IgG and F(ab)2 and purity of IgG were determined by UVspectrophotometer which showed Ab1 concentration 1 mg/ml. Molecular weight was estimated by sodiumdodecyl sulfate- polyacrilamide gel electrophoresis (SDS-PAGE). Ab2 was produced by immunization ofrabbit with Ab1. The first immunization was carried out by subcutaneous injection with 500 ?g of Ab1emulsified in Complete Freund Adjuvant. The immunization was repeated with the same dose of Ab1emulsified in Incomplete Freund Adjuvan at 1 week intervals. One week after the second immunization,rabbit’s serum was harvested and IgG was purified using the Montage Antibody purification kit & spincolumn with Prosep A media (Millipore). The rabbit IgG, called Ab2, was an anti-idiotypic antibody againstAIV-H5N1. In AGPT, a precipitation line appeared between Ab1 and Ab2. A partial reaction appearedbetween Ab2 and the AI H5N1 antigen was also detected. The results indicated that Ab2 is a possiblecandidate of imunogen for protection against an AI virus H5N1 infection.

Abstract

A severe and extensive intestine damages can occur in dogs suffering from volvulus , strangulation,intersuception and neoplasia. Extensive enterectomy is the most common medical treatment for dogssuffering from such disorder. A study was therefore conducted to compare the serum electrolyte level ofextensively enterectomyzed dogs after being treated with different level (0%, 0.05% and 0.5 mg/bodyweight) of lactoferin. As many as 9 dogs at 3-4 moth-old and with the body weights ranging from 4-5 kgwere used. After being enterectomized to the extent of 75% out of the total length of the intestine, the dogswere divided randomly into 3 groups (I, II and III) each of which consisted of 3 dogs. The three groups weretreated for 30 days respectively with 0.0 mg/body weight, 0.05mg/kg body weigth and 0.5mg/kg bodyweight of lactoferin. The electrolyte levels (Na, K and Cl ion) of dogs were determined at days 1, 15 and 30of during the treatment. The data collected from this study were analysed by Analysys of Variance (Anova)proceded by Duncan Multiple Range test (DMRT). The result showed that at days 15 and 30 of thetreatment, the electrolyte levels of the dogs with lactoferin (0.05 and 0.5 mg/body weight) were significantlyhigher than in dogs without lactoferin (0mg/bodyweight). The level of Na ion of dogs with 0,5, 0,05, dan 0mg/kg bw lactoferin were 143, 143,4, dan 141,7 mEq/L respectively at day 15 and 147, 150, dan 137,7mEq/L respectively at day 30. The levels of K ion for those dogs were 5,17, 4,97, dan 3,83 mEq/Lrespectively at day 15 and 30: 5,1, 5,13, 3,73 mEq/L respectively at day 30. Meanwhile, their Cl levelswere 113,7, 114,3, 104 mEq/L respectively at day 15 and 115,3, 117,3, dan 91,3 mEq/L respectively atday 30. Hasil penelitian ini menunjukkan bahwa memperian lactoferin memperbaiki kadar elektrolitserum anjing yang dienterektomi secara ekstensif. It was evidence that lactoferin treatment can improvedthe electrolyte profiles of extensively enterotomized dogs.

Abstract

Mycobacterium avium subspesies paratuberculosis (MAP) is a thermal tolerant bacteria. The presenceof these bacteria in pasteurized dairy milk is associated with infectious bowel disease in human known asCrohn’s disease. The aim of this study was to detect MAP in pasteurized dairy milk sold in Bogor. Fourtytwo samples of plain flavoured milk (180–250 ml) from 7 producers were bought from supermarkets inBogor. The presence of MAP was detected by isolation and conventional polymerase chain reaction (PCR)using IS 900 and F57. Bacterial isolation were done by Herrold’s egg yolk medium with mycobactine J(HEYMj) and without mycobactin J (HEYM) and incubated at 37°C for 20 weeks. The DNA extraction ofall pasteurized dairy milk samples were conducted by DNeasy® Tissue Kit. Amplification conditionsfor PCR were: 1 cycle at 94°C for 10 minutes, 40 cycles at 94°C for 1 minute, 58°C for 1 minute, and72°C for 3 minutes, and 1 cycle at 72°C for 7 minutes. After 20 weeks of incubation, there were no sign ofMAP which grew in all isolation mediums. The PCR IS 900 and F57 did not detect the DNA band of thetarget. In the conclusion, there was no MAP detected in pasteurized dairy milk sold in Bogor.
